Four papers in Cell today from my lab and collaborators identify targetable pathways—e.g. lipid metabolism, 3D genomic disruption, inflammatory dysregulation of microglia—with a major roles in Alzheimer’s disease. The teams analyzed transcriptional and epigenomic patterns in more than 400 postmortem AD and non-AD human brains and then tested leading hypotheses at the bench. We also found that specific inhibitory neurons are key to retaining cognitive function amid pathology. Importantly, unrepaired DNA damage in neurons can lead to mosaic gene fusion and disruption of 3D genome structure that impairs neuronal gene expression.
